2D in vitro models to assess target organ toxicity potential of chemicals suffer from major limitations arising from lack of maintenance of physiological and chemical metabolism competence. 3D cell cultures have been receiving increasing attention as they permit the incorporation of 3D architecture, multiple cell types, flow and oxygen gradients, thereby better recapitulating an in vivo microenvironment. As these models have become more widely adopted, there has been corresponding improvements in high tissue-specific functionality, long-term viability, robustness, endpoint analysis, high-throughput screening and imaging analysis of these systems. This course will provide a review of different 3D microtissue and fluid-flow bioreactors used in toxicology and drug screening with a concise overview of the technologies, advantages and cost. Application examples of 3D cell-based systems to skin, heart and liver will be presented, in addition to a comparison of scaffold-free and scaffold-based methods.

Drug safety in human patients is the ultimate goal of drug safety scientists. Because limitations of patient populations studied during drug development and uncertainties related to potential drug-drug interactions or rare or idiosyncratic adverse reactions, it is important to pay careful attention to human safety to achieve a better understanding of the safety of drugs on the market used in a wider patient population, for long treatment duration and on various co-medication compared to the population included in the approval package. Human safety plays an important role for assessing drug safety profile in daily conditions of use, which includes adherence to recommended modalities of therapeutic use (pharmacovigilance) as well as exposures deriving from accidental or intentional acute poisonings (clinical toxicology and toxicovigilance), misuse or abuse of therapeutic agents (pharmacodependence) or unintended indications.
